Using markers to assess recombination

This figure shows the profile of parents 1 and 2 assayed with CAPs marker X. The first lane is a size standard. There is a clear polymorphism between the two parents. If both parents showed the same band (i.e. they were monomorphic for this marker), this would not be a useful marker.

Note: CAPS markers identify polymorphisms by digesting the PCR products with a restriction enzyme. The parent 2 profile consists of two restriction fragments, but the parent 1 amplified segment contains no restriction site, and thus is not cut by this particular enzyme.
